Can I use this with a Canon 500D?

hello, the answer is yes as long as you have the standard tripod thread at the bottom of your camera,there is not a problem. i use it on my canon 50D and my two other fujifilm cameras, so they are very versatile. i hope this helps.

Will this fit non-Manfrotto monopod?

yes, i use a benro monopod and is great, the tilt head has both 3/8ths and 1/4inch fit female socket, might need the 1/4" to 3/8" Camera Convert Screw Adapter for Tripod Monopod depending what size your monopod male screw is, adapter`s are very cheap to buy on amazon, normally get 3 or 4 for around £2, just make sure it screws right in to the tripod head and should be flush fit for stable mounting.

I have been given a manfrotto monopod. However, the male thread on it is too big for my cameras.Does this device solve that?

I agree this is a bit confusing as it's not clear when you are getting the monopod exactly what the tread sizes are. If your monopod is too large for the camera you have a 3/8" thread. The monopod head (which is an excellent bit of kit) usually has a 3/8" thread in the bottom bit (check this when you order). The detachable top plate should have a 1/4" male stud to fit your camera OK.
