Do You need these to play the game?

Nope! They are not needed for 100% completion. They will give you some outfits when you use them the first time but you are able to buy/earn the outfits later on in the game without needing the Amiibo. They do provide some other useful benefits (e.g. the Bowser one will help you find kingdom-specific coins), but it's not necessary to play the game.
